Some imperfect specimens deposited in the herbarium HIRO, which were difficult to identify to species level based on morphology, were identified using DNA barcoding combined with molecular phylogenetic analyses. Based on sequences of the chloroplast ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit (cp rbcL) gene and the nuclear ribosomal internal transcribed spacer (nr ITS) region, Ludwigia ovalis Miq. (Onagraceae) and Juncus wallichianus Laharpe (Juncaceae) were identified from herbarium specimens collected in Miyajima Island, Hiroshima Prefecture, Honshu, southwestern Japan. L. ovalis was formerly regarded as extinct on the island. According to locality information on the specimens, these species grew in a small artificial pond in a small valley in forest dominated by Abies firma Siebold & Zucc. and Symplocos glauca (Thunb.) Koidz. The results demonstrate that DNA barcoding combined with molecular phylogenetic analyses are useful tools for species level identification of imperfect specimens.
